Salmonella infection is a battle between good and bad bacteria in the gut

June 26, 2013 ? The blockbuster battles between good and evil are not just on the big screen this summer. A new study that examined food poisoning infection as-it-happens in mice revealed harmful bacteria, such as a common type of Salmonella, takes over beneficial bacteria within the gut amid previously unseen changes to the gut environment. The results provide new insights into the course of infection and could lead to better prevention or new treatments.

"We're trying to tease apart a largely unknown area of biology," said systems biologist Josh Adkins and team lead at the Department of Energy's Pacific Northwest National Laboratory. "Infection changes the populations of bacteria in the gut with resulting inflammation. We want to understand the interplay between these events."

Out this week in PLOS ONE, the study shows that Salmonella Typhimurium might use the sugar fucose either as a sign that it has found a good place to reproduce or use fucose to sustain itself during infection, or both. This was the first time researchers saw fucose as an important player during Salmonella infection.

"We were taken completely by surprise with the fucose results," said Adkins. They also saw other sugars that normally are eaten by resident bacteria going untouched. "By knowing what the bacteria eat, we can try to promote the good bacteria and throw off the battle."

The Mice

Food poisoning caused by Salmonella bacteria hits more than 40,000 people every year. One of the common types that infect people, Salmonella Typhimurium, doesn't usually get mice sick, so Adkins and colleagues used mice uniquely sensitive to Salmonella infection. After infecting mice with the disease-causing bacteria orally, the researchers could follow the course of the illness by analyzing what came out of the other end of the mice.

"In most studies, researchers clear out the resident bacteria with antibiotics before introducing infectious bacteria," said microbiologist Brooke Deatherage Kaiser. "In this study, we could watch Salmonella knock out the commensal organisms and then watch them come back. Following the interactions through time is not something we've been able to do before."

The story they put together shows how Salmonella usurps microbes that normally populate the gut. Known as commensal bacteria, resident bugs perform important functions such as breaking down carbohydrates and sugars that people and mice can't. Using advanced instruments and techniques, the researchers identified which populations of bacteria dominated as infection progressed and mice recovered, as well as changes in the gastrointestinal tract, such as the presence of inflammation and available nutrients. Some of the experiments were performed in EMSL, the DOE's Environmental Molecular Sciences Laboratory on PNNL's campus.

The Sugars

While many events the team witnessed were expected, such as infection causing inflammation in the gut, some were not. One unexpected change was in the kinds of sugars available for bacteria to eat. A handful of sugars that good bacteria normally chow down on lay around the gut untouched.

This stockpile of unusual sugars likely occurred because the good bacteria had, by that point, been overtaken by Salmonella and another bacterial variety, Enterococci. Enteroccoci are normally found in the gut, but can take advantage of opportunities to overgrow their welcome.

Unexpectedly, several lines of evidence suggested that Salmonella might use the sugar fucose as a food source. This study showed that the bacteria produced proteins that specifically help it digest fucose, which was the first time these researchers observed fucose proteins during Salmonella infection.

Although additional research will be needed to flesh out the role of fucose in the infectious cycle of Salmonella Typhimurium, this observation may help to control or prevent gastrointestinal infection in the future by a better understanding of nutrient sources and signals in the gut.

Overall, the study allowed the PNNL researchers to follow the rise and fall of the infecting bacteria, the fall and rise during recovery of the commensal bacteria, and changes to the gut as the mice fended off the infection. Future research will focus on what happens in other areas of the intestine to get a handle on the difference between the type of illness this study represented, acute gastrointestinal disease, and more systemic infection.

It’s a pretty simple game, so we’ll see if that ? and the vanity aspect of getting to upload your own photos ? will be enough to make it as addictive as Litvak is hoping.

Assad lifts Syrian wages after conflict hikes prices

BEIRUT (Reuters) - President Bashar al-Assad ordered pay rises for Syrian soldiers and government employees on Saturday to address steep increases in the cost of living caused by the two-year uprising against his rule.

State media said the pay hikes would raise monthly wages by 40 percent for the first 10,000 Syrian pounds ($50), tapering to 5 percent for monthly pay above 30,000 pounds. Retired state employees would get more modest rises, it said.

Sharp falls in the Syrian pound have driven up the price of many goods in recent days. The currency, which stood at 47 to the dollar at the start of the conflict in March 2011, has tumbled in value to below 200.

Inflation stands at 50 percent but economists warn that sustained falls in the currency could fuel hyperinflation.

Egypt cuts diplomatic ties with Damascus, backs no-fly zone

On Saturday, Egyptian President Mohamed Mursi announced Egypt was cutting all diplomatic ties with Syrian President Bashar al-Assad's government and backing a no-fly zone over Syria. Mursi also called on Hezbollah to leave Syria.

Addressing a rally called by Sunni Muslim clerics in Cairo, the Sunni Islamist head of state also warned Assad's ally, the Iranian-backed Lebanese Shi'ite militia Hezbollah, to pull back from fighting in?Syria.

"Hezbollah must leave?Syria. These are serious words," said Mursi, whose country hosted a conference of Sunni clerics this week who issued a call for holy war against?Damascus.

"There is no space or place for Hezbollah in?Syria," Mursi said.

The rally underscored the region's deepening sectarian rift. A cleric who spoke before Mursi described Shi'ites as heretics, infidels, oppressors and polytheists.

It was also a show of support for Mursi as his opponents mobilise for protests to demand early presidential elections.

Mursi waved Syrian and Egyptian flags as he entered the auditorium packed with 20,000 supporters. The crowd chanted: "From the free revolutionaries of?Egypt: We will stamp on you, Bashar!"

Mursi, a?Muslim Brotherhood?politician, steered clear of direct references to Shi'ites and Iran but in a partial allusion to?Tehran, he accused states in the region and beyond of feeding "a campaign of extermination and planned ethnic cleansing" in?Syria.

"We decided today to entirely break off relations with?Syria?and with the current Syrian regime," he said. He also urged world powers not to hesitate to enforce a no-fly zone over?Syria.

Western diplomats said on Friday that Washington was considering a limited no-fly zone over parts of?Syria, but the?White House?said later that the?United States?had no national interest in pursuing that option.

Russia, an ally of Assad and fierce opponent of outside military intervention in?Syria, said any attempt to impose a no-fly zone using F-16 fighter jets and Patriots based in?Jordan?would be illegal.

Mursi said he was organising an urgent summit of Arab and other Islamic states to discuss the situation in?Syria, where the?United States?has in recent days decided to take steps to arm the rebels.

Egypt's U.S.-funded and -trained army is among the most powerful in the?Middle East. There has been no suggestion, however, that?Egypt, a country steeped in poverty, should get involved in the fighting in?Syria.

WARNS AGAINST VIOLENCE

Mursi said: "The Egyptian people supports the struggle of the Syrian people, materially and morally, and?Egypt, its nation, leadership ... and army, will not abandon the Syrian people until it achieves its rights and dignity."

The Brotherhood has joined calls this week from Sunni Muslim religious organisations for jihad against Assad and his Shi'ite allies.

Egypt?has not taken an active role in arming the Syrian rebels, but an aide to Mursi said this week that Cairo would not stand in the way of Egyptians who wanted to fight in?Syria.

It marked Mursi's second combative foreign policy speech in less than a week. On Monday, he said?Egypt?would keep "all options open" for dealing with a dispute with?Ethiopia?over a giant dam it is building on the Nile, though he said Cairo did not want war and stressed it would work diplomatically.

Mursi's liberal and leftist opponents are mobilising for mass protests on June 30, the anniversary of Mursi coming to office, fuelling fears of possible further violence.

(AP) ? After months of threatening to wage a nuclear war, North Korea did an about-face Sunday and issued a surprise proposal to the United States, its No. 1 enemy: Let's talk.

But the invitation from North Korea's National Defense Commission, the powerful governing body led by leader Kim Jong Un, comes with caveats: No preconditions and no demands that Pyongyang give up its prized nuclear assets unless Washington is willing to do the same ? ground rules that make it hard for the Americans to accept.

Washington responded by saying that it is open to talks ? but only if North Korea shows it will comply with U.N. Security Council resolutions and live up to its international obligations.

"As we have made clear, our desire is to have credible negotiations with the North Koreans, but those talks must involve North Korea living up to its obligations to the world, including compliance with U.N. Security Council resolutions, and ultimately result in denuclearization," U.S. National Security Council spokeswoman Caitlin Hayden said in a statement. "We will judge North Korea by its actions, and not its words and look forward to seeing steps that show North Korea is ready to abide by its commitments and obligations."

North Korea's call for "senior-level" talks between the Korean War foes signals a shift in policy in Pyongyang after months of acrimony.

Pyongyang ramped up the anti-American rhetoric early this year after its launch of a long-range rocket in December and a nuclear test in February drew tightened U.N. and U.S. sanctions. Posters went up across the North Korean capital calling on citizens to "wipe away the American imperialist aggressors," slogans that hadn't been seen on city streets in years.

The U.S. and ally South Korea countered the provocations and threats by stepping up annual springtime military exercises, which prompted North Korea to warn of a "nuclear war" on the Korean Peninsula.

But as tensions began subsiding in May and June, Pyongyang began making tentative, if unsuccessful, overtures to re-establish dialogue with Seoul and Washington.

Earlier this month, it proposed high-level talks with South Korea ? the first in six years. But plans for two days of meetings last week in Seoul dramatically fell apart even before they began amid bickering over who would lead the two delegations.

Meanwhile, the virulent anti-American billboards plastered across the city were taken down. And on Sunday, as scores of people fanned out across Pyongyang to help carry out the latest urban renewal projects in the capital ? landscaping and construction ? the National Defense Commission issued a statement through state media proposing talks with the U.S. to ease tensions and discuss a peace treaty formally ending the Korean War.

North Korea fought against U.S.-led United Nations and South Korean troops during the three-year Korean War in the early 1950s, and Pyongyang does not have diplomatic relations with either government. The Korean Peninsula remains divided by a heavily fortified border.

Reunifying the peninsula was a major goal of North Korea's two late leaders, Kim Il Sung and Kim Jong Il, and is a legacy inherited by current leader Kim Jong Un. North Korea is expected to draw attention to Korea's division in the weeks leading up to the 60th anniversary in July marking the close of the Korean conflict, which ended in an armistice. A peace treaty has never been signed formally ending the war.

Across Pyongyang, signboards at construction sites are marked with a countdown to July 27, giving laborers a deadline for retiling the roof of the People's Palace of Culture, renovating the Korean War museum, and planting trees and grass meant to beautify the city for the milestone anniversary.

For the nation's leaders, July 27 may well be their deadline for drawing the United States to the negotiating table to discuss a peace treaty.

But for Washington, there will be no talks just for talks' sake, officials say.

Speaking on CBS television's "Face the Nation" show Sunday, President Barack Obama's chief of staff, Denis McDonough, said Washington has been "quite clear" that officials support dialogue and have engaged Pyongyang in talks in the past.

But "those talks have to be real. They have to be based on them living up to their obligations, to include on proliferation, on nuclear weapons, on smuggling and other things," he said. "And so we'll judge them by their actions, not by the nice words that we heard yesterday."

He said smooth talk will not help Pyongyang evade U.N. sanctions supported by Moscow and Beijing, North Korea's two traditional allies. U.N. Security Council resolutions ban North Korea from developing its nuclear and ballistic missile programs.

Earlier this year, Kim Jong Un enshrined the drive to build a nuclear arsenal, as well as expand the economy, in North Korea's constitution. Pyongyang, estimated to have a handful of crude nuclear devices, says it needs to build atomic weapons to defend itself against what it sees as a U.S. nuclear threat in Korea and the region.

The National Defense Commission reiterated its refusal to give up its nuclear ambitions until the entire Korean Peninsula is free of nuclear weapons, a spokesman said in a statement carried by the Korean Central News Agency.

"The denuclearization of the Korean Peninsula does not only mean 'dismantling the nuclear weapons of the North'" but also should involve "denuclearizing the whole peninsula, including South Korea, and aims at totally ending the U.S. nuclear threats" to North Korea, the spokesman said.

The U.S. denies having nuclear bombs in South Korea, saying they were removed in 1991. However, the U.S. military keeps nuclear submarines in the region and has deployed them for military exercises with South Korea.

After blaming Washington for raising tensions by imposing "gangster-like sanctions" on North Korea, the spokesman called on the U.S. to propose a venue and date for talks ? but warned against setting preconditions.

Washington has been burned in the past by efforts to reach out to Pyongyang.

Months of behind-the-scenes negotiations yielded a significant food-for-disarmament deal in February 2012, but that was scuttled by a failed North Korean long-range rocket launch just weeks later.

How Talking To Your Computer Can Improve Your Social Skills

People with a fear of social situations are often labeled as loners and ostracized, but in reality social phobias are incredibly common. For example, how many of us get anxious about speaking in front of a large crowd? And to help people over come these fears, researchers at MIT have developed an interactive program that coaches people through social interactions, which boosts their confidence.

The MACH?or My Automated Conversation coacH?software uses a computer generated onscreen persona that people with social phobias learn to interact with. A video camera constantly monitors the person's movements and facial expressions, while behavior analysis software determines how well they did in a conversation based on eye contact and reactions.

Feedback is provided on how the user can improve their social interactions, and since there's no fear of the on-screen simulation making fun of them, or reacting awkwardly to something they've said, it helps build and reinforce the confidence needed to tackle real world social interactions with ease.

(AP) ? The headliners usually get all the ink and this year's group at the Bonnaroo Music & Arts Festival is definitely ink-worthy: Paul McCartney, Mumford & Sons and Tom Petty.

It's the down-list acts, though, that make this a particularly strong Bonnaroo lineup with a number of must-see acts you ought to check out before they're so big you'll be standing at the back of the crowd watching them on the video screens.

Here are 10 to see:

Alt-J: This Mercury Prize-winning quartet plays angular pop songs that are oddly intoxicating. Like fellow Brits McCartney and Mumford, the group has a gift for sugary songs that are impossible to resist.

Action Bronson: The burly, bearded rapper from Queens is poised to release his major-label debut later this year and has been whipping up a frenzy in London before returning to the states for Bonnaroo. Expect stage diving, East Coast harmonics and lots of naughty humor.

Charli XCX: The alternapop princess has had the bloggers buzzing for a couple of years. Now she's attached to a worldwide hit ? she features on Icona Pop's "I Love It" ? and has the highest profile of her career coming into Manchester.

Father John Misty: Former Fleet Foxes drummer Josh Tillman left the band and released his first solo album under this new moniker after several releases as J. Tillman. "Fear Fun" was on many year-end lists and it will be interesting to see how far Tillman's mostly hushed folk-rock will carry at Bonnaroo.

HAIM: Los Angeles-based sisters Este, Danielle and Alana Haim form a girl group for the 21st century, mixing lush vocal harmonies with high energy, beat-oriented grooves that have won over a lot influential fans. They're managed by Roc Nation, recently collaborated with Diplo and Kid Cudi, and are working on a debut album.

Jason Isbell: The Alabama-raised, Nashville-based singer-songwriter is the Americana community's cause celeb du jour. He releases his new album "Southeastern" this week, just in time to take his brand of Southern rock to the masses.

Japandroids: Just when you think the two-man band thing has run its course, up pops Vancouver-based Japandroids, a band guaranteed to generate more decibels per band member than any other on the farm. Their soaring, anthemic rock is perfect for Bonnaroo.

Kacey Musgraves: Bonnaroo has had its share of edgy country acts over the years and Musgraves keeps the tradition rolling. This champion of Nashville songwriters has the off-kilter, left-leaning world view that fits right in at the festival.

Portugal. The Man: This Portland-based band of spacey rockers has joined with producer Danger Mouse on its fun new album, "Evil Friends." Fans at Bonnaroo will be hearing the new music for the first time. There will be buzz.

Tame Impala: Australian rocker Kevin Parker is the premiere purveyor of freaky, fuzzed-out psychedelic rock at the moment. Last year's "Lonerism" was one of rock's most praised albums and Bonnaroo could be a defining moment.

NASA astronaut Chris Cassidy recently demonstrated the fine art of shaving your head in space. It's more complicated than you might think.

Shaving your head is the simplest of haircuts here on Earth, but it presents some special challenges in space, as NASA astronaut Chris Cassidy recently demonstrated.

Cassidy shaved his head in orbit late last month to welcome chrome-domed Italian astronaut Luca Parmitano, who arrived at the?International Space Station?on May 28 along with NASA's Karen Nyberg and Russian cosmonaut Fyodor Yurchikhin.

"I'm going to go out on a limb here and get the full-on Luca Parmitano look for the hatch-opening ceremony," Cassidy said in a NASA?video showing the head-shaving process. "So please join me [at] Chris' Barber Shop."

Cassidy held a hair trimmer up to the camera, then connected it to a vacuum cleaner using a long tube.

"Otherwise, the hair would get all over the place," he explained.

Then Cassidy commenced buzzing his hair, which was pretty short to begin with. He stopped halfway through the process and smiled at the camera, showing off a temporary mohawk.

"I don't think I've looked like this since Plebe Summer," the former Navy SEAL said as he shaved off the mohawk, referring to the training program the United States Naval Academy puts all of its incoming freshmen through.

After finishing with the clippers, Cassidy reached for a razor to give himself Parmitano's sleek, smooth-pated look.

"I wonder if he does this every day," Cassidy said before wiping his newly shorn scalp down with a small white towel. "It kinda hurts."

While head-shaving may not have been filmed in space before, Canadian astronaut Chris Hadfield demonstrated how he?shaved his face in microgravity?in April.

Cassidy and Russian cosmonauts Alexander Misurkin and Pavel Vinogradov launched toward the orbiting lab aboard a Soyuz spacecraft on March 28. Along with Parmitano, Nyberg and Yurchikhin, they make up the crew of the space station's current Expedition 36.

Cassidy, Misurkin and Vinogradov are slated to return to Earth in September. Their three crewmates will remain in orbit a few more months, coming home in November.

South Africa: Mandela taken to hospital

JOHANNESBURG (AP) ? Former South African President Nelson Mandela is in "serious but stable" condition after being taken to a hospital to be treated for a lung infection, the government said Saturday, prompting an outpouring of concern from admirers of a man who helped to end white racist rule.

Mandela, who is 94 years old, was treated in a hospital several times in recent months, with the last discharge coming on April 6 after doctors drained fluid from his lung area after diagnosing him with pneumonia. He has been particularly vulnerable to respiratory problems since contracting tuberculosis during his 27-year imprisonment under apartheid.

"During the past few days, former President Nelson Mandela has had a recurrence of lung infection," said a statement from the office of President Jacob Zuma. "This morning at about 1.30a.m., his condition deteriorated and he was transferred to a Pretoria hospital."

It said Mandela was receiving expert medical care and "doctors are doing everything possible to make him better and comfortable."

Zuma wished Mandela a quick recovery on behalf of the government and the nation and requested that the media and the public respect the privacy of the former leader and his family, the statement said.

The African National Congress, the ruling party that has dominated politics in South Africa since the end of apartheid, said it hoped Mandela, known affectionately by his clan name Madiba, would get better soon.

"We will keep President Mandela and his family in our thoughts and prayers at this time and call upon South Africans and the peoples of the globe to do the same for our beloved statesman and icon, Madiba," the party said in a statement.

On April 29, state television broadcast footage of a visit by Zuma and other ANC leaders to Mandela at his Johannesburg home. Zuma said at the time that Mandela was in good shape, but the footage - the first public images of Mandela in nearly a year - showed him silent and unresponsive, even when Zuma tried to hold his hand.

South Africans expressed hope that Mandela would recover from his latest setback.

"He is going to survive," said Willie Mokoena, a gardener in Johannesburg. "He's a strong man."

Another city resident, Martha Mawela, said she thought the former president would recover because: "Everybody loves Mandela."

Mandela was robust during his decades as a public figure, endowed with charisma, a powerful memory and an extraordinary talent for articulating the aspirations of his people and winning over many of those who opposed him.

In recent years, however, he has become more frail and last made a public appearance at the 2010 World Cup soccer tournament, where he didn't deliver an address and was bundled against the cold.

In another recent hospitalization, Mandela was treated for a lung infection and had a procedure to remove gallstones in December. In March, he spent a night in a hospital for what authorities said was a scheduled medical test.

Hearing Aids Can Serve a Second Purpose As Wireless Speakers

?Never in my audiology career has something so simple helped so many people at so little cost.?

?I can?t stop smiling.?. . . I could understand every word. . . .? What an overwhelming experience.?

?I am not exaggerating when I tell you that at EVERY [theatrical] performance we get big thank you?s.? We have subscribers who are returning and telling their friends.?

These reports?from an audiologist, a person with hearing loss, and the business manager of an 882-seat Chicago theatre?aptly describe my response when first experiencing the hearing assistance technology to which they each refer.? As I sat, unable to understand the words reverberating off the ancient stone walls of Scotland?s Iona Abbey, my wife noticed a hearing assistance sign with a ?T? and nudged me to turn on the ?telecoils? in my new aids.? The instant result was a stunningly clear voice, speaking from the center of my head.? I was on the verge of tears.

In our subsequent UK sojourns, I have experienced the spread of this ?hearing loop??technology?to auditoriums, churches and cathedrals, and to tens of thousands of transient venues, including ticket and post office windows, and all London taxis.? Although specific applications need professional design, the gist of the technology is simple:? an amplifier picks up sound from a PA system and transforms it into a magnetic signal sent by a wire loop surrounding an audience.? The telecoil?an inexpensive magnetic sensor in most of today?s new hearing aids and all new cochlear implants?receives this signal, enabling the hearing instrument to become an in-the-ear speaker that broadcasts sound appropriate to each user?s needs.? And I bet you didn?t know this:? all U.S. landline phones, and select mobile phones (including my new iPhone5), also are ?hearing aid compatible.? That means they can similarly transmit improved sound via magnetic communication with telecoil-equipped hearing instruments.

Back home, I installed hearing loops in my home and office.? Voila!, my TV and phone now broadcast wonderfully clear sound through my aids (and binaural phone listening is vastly superior to one-eared listening).

Other modern wireless technologies also connect hearing aid wearers to their TVs and phones.? When connected to a TV or stereo, a transmitter can ?stream?? audio directly to one?s hearing aids.? When synced with one?s phone, conversation can be broadcast to both hearing aids.? Bluetooth-enabled hearing aids can receive sound from a host of Bluetooth devices, including phone and computer.

So, people often wonder, might Bluetooth or other streaming wireless technologies enable assistive listening in public venues?and without the expense of a running a loop wire?

Alas, each hearing aid company offers a different proprietary wireless technology.? The one universal wireless receiver?which anyone can use, no matter what their hearing aid brand or what country they are in?is the humble telecoil (a part that costs hearing aid companies about $2).? Moreover, unlike Bluetooth?s battery-sucking demands, the telecoil requires zero power. With today?s popular ?open-fitting? hearing aids, a person may hear both immediate sound from a TV and slightly delayed Bluetooth sound, creating an annoying echo.? And unlike Bluetooth and other wireless technologies that serve only small areas, hearing loops work in places both small and big.

Indeed, the accelerating move to hearing loops in the USA?sparked by local and national hearing loss associations and now supported by a new cottage industry of hearing loop vendors and installers?has led to thousands of newly looped venues.? Some venues are small, such as New York City?s 450 subway booths and all its future taxis.? Some are bigger, such as auditoriums and worship places.? And some are huge, such as airports and Michigan State University?s basketball arena.

Are hearing loops the final word in assistive listening?? Likely not.? But the challenge for hearing technologists is to make any alternative technology similarly

  • simple for people of all ages to operate (without needing to pair and charge special equipment),
  • affordable, without adding to the cost of already-expensive hearing instruments,
  • available with nearly all hearing instruments,
  • energy efficient,
  • scalable, with applications in public spaces both small and vast, and
  • universal, with the same signal serving everyone, no matter their location or hearing instrument manufacturer.

Given a future with a universal wireless receiver (for now, a telecoil) in virtually every hearing instrument, and given the continuing spread of hearing aid compatible assistive listening, we can envision a better future for Americans with hearing loss?a future in which hearing aids will serve as customized, wireless speakers in all sorts of public venues.? That, methinks, would be a future where doubly useful hearing aids for challenged ears would be as commonplace as glasses for challenged eyes.

Study suggests fathers should ask kids: 'Am I the dad you need me to be?'

June 5, 2013 ? As Father's Day draws near, psychologist Jeff Cookston says dads should ask their children for a little more feedback than they might get with the yearly greeting card.

Just being a good parent may not be good enough, said Cookston, professor of psychology at San Francisco State University, who has studied fatherhood extensively. "There's a need for fathers to sometimes say to their kids, 'How am I doing? Am I the dad you need me to be?'"

"Kids are actively trying to make sense of the parenting they receive," he explained, "and the meaning that children take from the parenting may be as important, or more important, than the behavior of the parents."

"I don't think a lot of parents give these ideas about meaning much thought," Cookston said. "You may think that you're being a good parent by not being harsh on your kid, for instance, but your child may view that as 'you're not invested in me, you're not trying.'"

The meanings that adolescents in particular attach to their fathers' behavior can vary depending on the child's gender, ethnicity, and the presence of a stepfather in the child's life, Cookston and former SF State graduate student Andrea Finlay report in a new study published in the Journal of Family Issues. The study included children from California and Arizona.

The researchers examined how adolescents view their fathers' actions -- specifically, whether the teens attribute these actions to a dad's overall character or to his reaction in a particular situation. For instance, a daughter might believe her dad took her to the baseball game because he is a good father, or she might believe that he took her to the game because he likes to go to the game.

The study suggests that girls tend to believe that a father's "enduring aspects" are responsible for a dad's good deeds, while boys are more likely to think that dads do good depending on the situation. Mexican-American children are more likely than their European-American peers to think that good times with dad depend on the situation.

The reasons for these differences are not clear, Cookston and his colleagues say, although in the case of boys and girls, it may be that girls are socialized to interpret other people's behavior in a more positive light. In Mexican-American families, the process of adapting to U.S. culture may increase family conflict, leading children to have a less optimistic view of their fathers' good deeds.

Cookston has conducted extensive research on parenting and fatherhood, with a focus on how children from diverse cultural backgrounds respond to parenting and how children perceive and construct their relationships with their fathers. His research has shown that the relationship between father and child can have a significant impact on the child's tendencies toward depression and behavior problems.

Father's Day can be a good time for dads to rethink their relationship with their children, with a few tips that Cookston has gleaned from these studies:

Be sure to check in with your child. Dads may be surprised by the "filters" their children use to interpret their behavior, making it important for fathers to regularly ask about the relationship. "Fathers should ask, 'am I more or less than you need me to be?'," Cookston said, "and children -- particularly adolescents -- should be able to say, 'I need you to change course.'"

Show your emotional support. Dads provide everything from discipline to role modeling, but Cookston said it is the fathers who emphasize their emotional relationships with their children who have kids that are less likely to behave in aggressive and delinquent ways.

Don't be afraid to switch up your style. If you weren't always a warm and accepting father, it's not too late to become one, according to Cookston. "Parents can change, and kids can accept that. Parents need to be constantly adapting their parenting to the development and individual needs of the child."

Be a team player. Cookston's research focuses on dads, but his work with divorced families has taught him how valuable it is when parents work together as a team. Children are more likely to talk to parents about family relationships if they see that they agree on parenting decisions, he noted, and "parents play unique, additive roles in their children's lives."

Aim high as a dad. "We need to raise the bar for fatherhood. If a man is around and is a good provider and doesn't yell at his kids and goes to soccer games, we say that's enough," Cookston said. "But we need to expect more in terms of engagement, involvement and quality interaction."

Candidate drug provides benefit in spinal muscular atrophy animal models

June 4, 2013 ? In a new publication that appears in Human Molecular Genetics, the laboratory of Christine DiDonato, PhD reports on their pharmacological characterization of the drug RG3039, demonstrating that it can extend survival and improve function in two spinal muscular atrophy (SMA) mouse models. They have determined the minimum effective dose and drug action, thus contributing to dose selection and exposure estimates for the first studies with RG3039 in humans. As in cellular assays, the animal studies have shown that drug treatment leads to improvement in nuclear gem/Cajal body numbers in motor neurons.

Gem loss is a cellular hallmark of fibroblasts derived from SMA patients; gem numbers inversely correlate with SMA disease severity. In addition, the laboratory has shown improved functional outcomes, including treadmill walking and gait dynamics, in animals receiving the drug. The laboratory has been testing RG3039 in SMA mouse models with disease phenotypes ranging from mild to severe.

The collective results suggest that RG3039 positively modifies motor unit pathologies and dysfunction, and that it may have therapeutic benefit for SMA.

Spinal muscular atrophy (SMA) is a devastating hereditary disease that is a leading cause of infant and early childhood mortality. Motor neurons in the spinal cord are affected, resulting in muscle weakness and in many cases breathing problems. SMA is caused by insufficient levels of the survival motor neuron (SMN) protein; depending on the level of SMN protein, patients may have mild to severe forms of SMA. There is currently no cure for the disease.

The goal of early SMA drug discovery programs has been to identify small molecules that induce the SMN gene to produce sufficient levels of protein to improve motor neuron functioning in affected patients. A promising drug candidate is RG3039, which has been shown to increase nuclear gem/Cajal body numbers in cellular assays.

Says DiDonato: "We are very happy that our research was able to assist in the pharmacological characterization of RG3039 and contribute to dose selection and exposure estimates for the first studies with RG3039 in human subjects. How Wikipedia Would Have Worked in the 1980s

I'm so thankful I never used a computer in the 1980s because it looks mind-scrunchingly frustrating. I would probably hate technology because of it. So thank you to all the patient geniuses who took the time to use things like DOS for the rest of us. Even if today's tools existed back then, it would still be so much worse with yesterday's limitations.
